I'm thinking about putting Long Tube Headers on my LS1 Camaro. The only drivetrain upgrade other than SLP Lid.
It's my understanding I will need a tune adjustment after installing the LTs. Is it as simple as updating the fuel trims and timing, or is it more involved? Is it something I might be able to handle, or should I take it to a professional?

Kind of depends how much time you can spend on it
It is pretty straight forward with lots of tutorials on here.
Probably the most complicated thing will be to get your o2 sensors adjusted for the new location and different exhaust values.
With some basic steps you can have it safely driving pretty quick.
To dial everything to a "T" takes time and lots of logging.
Get a wideband oxygen sensor for the exhaust and it will make things easier.
Just adding long tube headers won't be that big of a task to tune. You can take your timing getting fueling and timing under control.
