Yes you can.
It will read whatever is on the computer at that given time. So if the vehicle was tuned previously with a Diablo or SCT tuner it would pull that file. Only stock vehicles will have stock calibrations in them.
Just note that not every tuning software will allow you to view and edit already modded files. Some of those other tuners will put locks on the computer so you will not be able to access them.
