Just as the title says, Will the original MPVI read and flash 2018 Challenger 6.4 stick shift, or do I HAVE to get the MPVI 2?

MPVI 1 will do it. Just keep in mind that regardless of MPVI 1 or MPVI 2, you'll need to send the PCM into HP Tuners to be unlocked (applicable to all 2015+ Chrysler/Jeep/Dodge), and will also need the Smart Access Cable (applicable to all 2018+ Chrysler/Jeep/Dodge).
